When reviewing analytical peptide testing, two words appear frequently: identity and purity. They are related—but they do not answer the same question.

Identity asks: “What molecule is present?”

Identity testing provides evidence that the material being examined corresponds to the expected compound.

Depending on the analytical objective, laboratories may use techniques such as mass spectrometry, chromatography and other complementary methods to establish identity.

Purity asks a different question

Purity testing evaluates impurities or the relative presence of the target material according to a specified analytical method.

For example, an HPLC report may provide a chromatographic area percentage based on the peaks detected under the particular test conditions.

Importantly, an HPLC area percentage should not automatically be interpreted as the percentage by mass of everything physically present in a vial.

Water, salts, counterions, residual solvents, excipients or compounds that are not detected under the particular analytical conditions may not be represented in the same way.

And what about quantity or assay?

Identity, purity and quantity are separate analytical questions.

A report might establish evidence about molecular identity, report chromatographic purity, measure the amount of a compound—or provide some combination of these.

One number should not automatically be substituted for another.

Purity: What does the chosen analytical method show about impurities or relative composition?

Quantity/assay: How much of the target material is measured?

Why multiple tests may be useful

No single analytical method necessarily answers every quality question. Complementary methods can provide different pieces of information about a sample.
